Impact of adding different concentrations of IGF-I and insulin to the semen extender on bull sperm quality post-cryopreservation

ABSTRACT This study aimed to evaluate the addition of different concentrations IGF-I and insulin egg yolk-based extender improve bovine semen cryopreservation. Two experiments were developed effects additives in two commercial extenders, Botubov® (Experiment 1) Triladyl® 2), both with same design. Three ejaculates from four bulls (n = 12) used. Each ejaculate was divided into seven equal fractions for dilution (60x106 spermatozoa/mL) following treatments: CON: only; IGF100: 100ng/mL; IGF200: 200ng/mL; INS150: 150µUI/mL; INS200: 200µUI/mL; ASS1: 100ng/mL + ASS2: 200ng/mL 200µUI/mL. Semen cryopreserved by an automated system. Post-thawed sperm evaluated regarding motility CASA (Computer-assisted analysis), membranes fluorescent probes (H342, PI, FITC-PSA JC-1). For extender, INS150 more efficient preserving total progressive motility, VCL, BCF, plasma mitochondrial membranes. A similar response seen when added membrane integrity potential. Thus, 150µUI/mL, regardless composition contributes better cryopreservation effects.
